Here we summarize recent advances in applying CRISPR/Cas9 methods in Lepidoptera, with a focus on providing practical advice on the entire process of genome editing from experimental design through to genotyping. We also describe successful targeted GFP knockins that we have achieved in butterflies. Finally, we provide a complete, detailed protocol for producing targeted long deletions in butterflies.
